Riken Beyond the Genome: Influenza, and Sugar Chains:DigInfo DigInfo - (http://movie.diginfo.tv) The Systems Glycan Biology Labs at RIKEN are exploring the role of glycans or sugar chains in interactions at the cellular level. These sugar chains are found on the outer membrane of proteins in the body and are essential in all interactions from embryonic growth to viral infections which is one of the focuses of the labs at RIKEN. Many other groups studying glycans currently are not examining the relationship of viruses and sugar chains but the Systems Glycan Biology Labs have three groups looking at the various aspects of viral infections at the cellular level. One of the ongoing studies is the transmission of the bird flu virus to humans. In a viral infection the viruses attach to the sugar chain and begin attacking the cell but certain viruses only attach to certain sugar chains. Originally the bird flu virus would not attach to human glycans and human flu viruses would not interact with bird cells. While these two viruses could not interact with the other species's sugar chains, both viruses are accepted by the sugar chains in pigs. While attached to the pig's cells the bird flu adapted and became able to attach to human cells. The researchers at RIKEN are researching medicines that will fight the flu virus by creating a decoy sugar chain that the virus would want to attach to and separating it from the real cell. This research is not only focusing on the flu virus. Through the research of sugar chains the Systems Glycan Biology Labs has found the cause of over 37 genetic diseases such as muscular dystrophy which progressively weakens and kills muscles in the body and generally affects many systems including the cardiovascular, digestive and nervous system. RIKEN Potential Drug for Alzheimer's : DigInfo DigInfo - (http://movie.diginfo.tv) A team from the Laboratory for Proteolytic Neuroscience at RIKEN is conducting various research on Alzheimer's disease especially in the study of somatostatin, a neuropeptide that inhibits the secretion of growth hormone and could lead to the discovery of new medicines for Alzheimer's disease. Alzheimer's disease is a cognitive dysfunction disorder that occurs when nerve cells are impaired and the brain becomes spongy. There are around 24 million Alzheimer patients worldwide. Amyloid β peptide has been identified as the pathogen that exhibits neurotoxicity and consists of 42 amino acids which are produced in the brain. Alzheimer's disease occurs when amyloidsβ-peptides accumulate in excess outside of the nerve cells. The Researchers at Riken also discovered that Neprilysin, which breaks up amyloidβ- peptide, diminishes with age and decreases even further upon the onset of Alzheimer's disease. In the laboratory, they treated an Alzheimer's disease model mouse with a gene therapy that boosted the activity of Neprilysin by 10 times, and as a result, were able to suppress the accumulation of the amyloid β- peptide. Somatostatin can act to reinforce memory retention, so it has the potential to be both a treatment and preventive drug for Alzheimer's disease. RIKEN is currently conducting bio-chemical and histological analysis of the brain using an Alzheimer's disease model mouse. RIKEN RI Beam Factory : DigInfo DigInfo - (http://movie.diginfo.tv) Riken researchers are exploring the most fundamental laws of physics in order to understand how the universe began using their RI beam factory which provides the world's most intense heavy-atomic-mass RI beams. Riken's RI Beam Factory can be used to research many aspects of physics from the origin of the universe, and the formation of heavy elements like uranium and gold, to the study of nuclear medicines for treating cancer and other diseases. In studying the beginning of the universe, the researchers are also exploring the periodic table of elements in particular heavy elements. The periodic table is organized by number of protons but certain elements have high atomic mass due to the extra neutrons in the nucleus. Elements which have an atomic mass higher than iron are considered heavy elements. This includes elements like gold and many radioactive elements such as Uranium. The origin or these elements is unclear but with the RI Beam Factory new heavy elements can be made. The Uranium ions are first generated in the ion source, and accelerated through a linear accelerator and the following four ring cyclotrons. This is the RI beam. The RI beam is accelerated to 70% the speed of light and then sent to the BigRIPS in which they collide with the target element and nuclear fission occurs. The beam is split in two and comprised of unstable nuclei then the mass and charge of each can be identified. The desired nuclei is then sent on to the RI Ring. Though the nuclei may have a short life span it is forced into an orbit and its mass can be measured accurately. Currently there are approximately 3,000 species of known nuclei and Riken aims to discover another 1,000 new nuclei. In biological research, the RI Beam Factory can be used to focus on various ions in living organisms that are used to transport metal ions in cells and send messages throughout the body. The researchers at the RI beam factory are collaborating with research centers worldwide to help explore various questions in physics such as how the first elements were created and the relationship of Supernovae, when a star explodes, to the origin of heavy elements. Riken Discovers Neural Response of Primary Visual and Auditory Cortexes Amplified by Focusing Attention : DigInfo DigInfo - http://movie.diginfo.tv Researchers at Riken's Human Brain Dynamics Laboratory have been successful in understanding how the brain's spatiotemporal response changes, depending on whether a person is paying attention or not, even when viewing or listening to the same stimuli. In our everyday surroundings, there is too much information for our brains to process all of it. So, it is believed that information that we focuse on is processed preferentially so that the limited processing capacity of the brain can be utilized efficiently. However, it had not been understood at what stage of sensory processing the brain selects information that is given attention, and in particular it had not been properly understood what sensory area is first affected as a result of focusing attention. The research team discovered that the initial neural response of the primary visual cortex (V1) and primary auditory cortex (A1), the first cortical areas of the brain receiving information about the stimuli, is amplified by attention. The results were obtained by analyzing magnetoencephalography, or MEG, signals and magnetic field tomography, or MFT. MEG measures the magnetic field outside of the brain, which is generated by neural electrical activity in the brain. MFT estimates the generators and represents them three-dimensionally as electrical current density distribution over all areas of the brain. In the experiment, the subject focused his attention on the area indicated on the screen in front of him. The team measured the difference in neural responses when pictures were shown in the attended area and outside the area. As a result, it was found that neural response elicited by the stimuli at the attended location is amplified in the primary visual cortex (V1) and primary auditory cortex (A1) as they first arrive there. Revealing the neural mechanism associated with focusing attention will help explain the higher brain function mechanism in humans. It is hoped this will lead to developing new treatment methods for mental illnesses such as dementia and schizophrenia in which there is damage to the higher brain function.
