El Norte - Parte I This movie is still without a doubt one of the best made about illegal immigration and specifically the push of refugees fleeing the violence in Central America. This movie is about hopes and aspirations of immigrants coming to the United States. Specifically, it deals with Guatemala where many indigenous people were killed by the military regimes in power there in the 1970s and 1980s. Many of the participants in the genocide have not been brought to justice and any recriminations are still met with violence. I remember watching it on PBS and in class during the 1980s. Gleevecs mechanism of Action The animation begins by introducing the Philadelphia Chromosome, the result of a reciprocal translocation between chromosomes 9 and 22. More specifically the breakpoint cluster region (BCR) of chromosome 22 is fused with part of the Abelson (ABL) gene on chromosome 9. The resulting BCR-ABL genetic domain now located within chromosome 22 and codes for a mutant tyrosine kinase also known as BCR-ABL. Under normal circumstances tyrosine kinase proteins respond to external cellular messaging proteins, and ultimately initiate a series of reactions that culminate in cellular replication. Conversely, BCR-ABL is constitutively active, meaning it does not require activation by the aforementioned cellular messaging proteins in order to stimulate cellular replication. This results in acceleration of cell division, an inhibition of DNA repair, overall genomic instability, and the fatal blast crisis characteristic of chronic myelogenous leukemia. The animation progresses to introduce Gleevec (imatinib), the first in a class of drugs that specifically target and competively inhibit the ATP binding site on BCR-ABL tyrosine kinase. This prevents the ABL domain from phosphorylating the tyrosine residue, and as a result preventing the proliferation of hematopoietic cells that express BCR-ABL. Therapy with imatinib results in a dramatic reduction of tumor clone cells and the occurrence of blast crisis', through targeted drug treatment which leaves health cells unscathed. Night Drive in Uptown Toronto and Markham Available in high quality. This is a video of me driving around the northern part of Toronto (more specifically, the borough of North York), and also in Markham, a suburb directly adjacent to Toronto from the northeast. The official border between the City of Toronto and its northern suburbs is Steeles Avenue, on which I drive in a major chunk of the video. I note it in the video when I first turn onto Steeles Avenue, specifically where it borders Vaughan in the west (another suburb), and then Markham in the east. The drive begins just before the end of the evening rush hour, and as a result traffic is quite heavy in the beginning. However, it quickly gets better. The video also features Pacific Mall, which is the largest Asian indoor mall in North America. Unfortunately we arrived just minutes away from closing time, although enough to make some last-minute purchases :) The drive itself may seem somewhat random in nature, but that mostly is a result of running some errands, picking up some friends, and doing some random driving while counting on the GPS to come up with a decent place to eat :) Also, many of the camera transitions are continuous in nature, and as a result it may look like I was just parking and then taking off right away, when in fact time has passed between the events. Fights average nearly an hour in length and often last more than two hours. Dogfights end when one of the dogs is no longer willing or able to continue. In addition to these dogfights, there are reports of an increase in unorganized, more spontaneous street fights in urban areas. The injuries inflicted and sustained by dogs participating in dogfights are frequently severe, even fatal. It was shot back-to-back with the video for "All Good Things (Come to an End)", the album's third single in Europe.
